    Cells respond to external stimuli by adopting complex transcriptional states, but most single-cell profiling captures only baseline conditions. Exploring functional cellular responses requires access to large combinatorial perturbation spaces, but single-cell genomics in standard macroscale arrayed formats is limited. We leverage StimDrop, a method that integrates antibody-based cell barcoding with droplet microfluidics to automate the generation of condition combinations, to explore the effects of sepsis-related cytokines on PBMC cell types.

    White Lab Knouse Labs

    The mammalian liver maintains a tightly regulated size “setpoint” and can restore appropriate mass following perturbations to liver size. However, the mechanisms governing liver size sensing and control remain unclear. Here, we integrated experimental and computational approaches to generate candidate regulators of this response and are using in vivo CRISPR-Cas12 screening to prioritize key mediators. Future studies will define upstream signals of these proteins, with potential to reveal therapeutic targets for modulating liver growth.
