A Quantitative Immunopeptidomic Platform for Identifying Treatment Associated Peptide Antigens

Abstract:

Combining targeted therapy and immunotherapy can extend cancer patient survival, but the optimal sequence of treatment remains poorly understood. Here, we investigate the immune mechanisms of adaptation to targeted therapies by quantifying how peptide MHC repertoires presented for immune recognition change with treatment. Our mass spectrometry based approach identifies antigens that increase their presentation in response to treatment, which we hypothesize can be leveraged for targeted immunotherapy to inform combination clinical trial design.
