Bathe LabDelivering therapeutics to the brain remains a major challenge. However, screening vector libraries in vivo often sacrifices spatial resolution for throughput, a major trade-off in an organ as structurally complex as the brain. We are developing DNA virus-like particles (D-VLPs) as an alternative non-viral vector for brain delivery. Using methods from spatial transcriptomics, we show that orthogonal barcoding of D-VLPs enables in situ tracking of library members with high cellular resolution, accelerating discovery of novel formulations.

Sun LabThroughout the animal kingdom, tissues and cell types follow shared and context-specific aging trajectories. Existing single-cell transcriptomic aging clocks show promise for evaluating aging interventions, but typically cover few biological contexts. Here, we develop universal single-cell transcriptomic aging clocks enabling comparison of intervention effects across biological scales, guiding rational design of combinatorial interventions targeting complementary aspects of aging.
