Barcoded DNA Nanostructure Libraries for Therapeutic Cargo Delivery

Abstract:

Targeted delivery remains a significant challenge for the clinical implementation of therapeutics. DNA nanostructures synthesized using scaffolded DNA origami offer a promising delivery vector with control over structure and spatial presentation of ligands, controllable immunogenicity, and low manufacturing costs. Large libraries of structurally unique nanostructures functionalized with targeting ligands are being fabricated to deliver therapeutic cargo. These libraries are identified using DNA barcodes, providing a high-throughput and scalable approach for engineering DNA nanostructures for therapeutic delivery.
