Abstract:
Enhancers are the primary regulators of mammalian gene expression; yet, the mechanisms underlying their interactions with gene promoters remain unclear.
Chromosome conformation capture (3C) methods enable 3D genome structure mapping, but have struggled to resolve enhancer-promoter interactions. Here, we
develop Region Capture Micro-C to generate the deepest reported 3D genome maps and uncover highly nested and focal enhancer-promoter interactions, which we term microcompartments and mechanistically distinguish from other genomic structures.
