Region Capture Micro-C Reveals Coalescence of Enhancers & Promoters into Nested Microcompartments

Abstract:

Enhancers are the primary regulators of mammalian gene expression; yet, the mechanisms underlying their interactions with gene promoters remain unclear. Chromosome conformation capture (3C) methods enable 3D genome structure mapping, but have struggled to resolve enhancer-promoter interactions. Here, we develop Region Capture Micro-C to generate the deepest reported 3D genome maps and uncover highly nested and focal enhancer-promoter interactions, which we term microcompartments and mechanistically distinguish from other genomic structures.

Dynamic Bottom-up Dissection of Chromatin Looping and Gene Regulation

Abstract:

Enhancers are the primary units of gene control and can activate promoters hundreds kb away. However, how enhancer-promoter (E-P) specificity is achieved remains poorly understood and is hindered by the redundancy of natural systems. To overcome this, we introduce a bottom-up SynBio approach via the engineering of synthetic topological associated domains (synTADs), as TADs are thought to control E-P specificity. We engineered synTADs ranging from 200 kb to 2.4 Mb in size, and tracked the 3D distance between synTAD boundaries in live cells. Next, we will insert an E-P pair aiming to elucidate the mechanistic rules that govern E-P specificity.
