Nuclear Mechanics in Cancer Metastasis

Abstract:

During cancer metastasis, cancer cells undergo large-scale deformations to traverse through cell-cell junctions in the endothelial wall of blood vessels. Deformation of the nucleus, which is much stiffer than the constitutive cell, may serve as a rate-limiting step in metastasis. Utilizing engineered in vitro vascularized tissue models and high resolution imaging, we mimic the physiological cancer microenvironment to shed light on the role of nuclear mechanics in cancer metastasis.
